No it doesn't have any actual effect - other than putting a bigger target on your back from the rest of the league - but it could be an indicator of overconfidence. These quotes are taken completely out of context. It's not as though these guys are being unprovoked and simply proclaiming that they're the best. Pace was asked how he believes the Jets defense stacks against the rest of the league and he says he thinks they're the best. What else should he say? I would be more concerned if he said that he thinks they're average. Geno said he thinks he'll be a top 5 QB this year which is exactly what he should think. Milliner said that he would never say that another CB is better than him. Good. Believing you're the best is the only way to become the best.
